Adrenergic signaling promotes the expansion of cancer stem-like cells of malignant peripheral nerve sheath tumors

Malignant peripheral nerve sheath tumor (MPNST), a highly malignant that arises in tissues, is known to be resistant radiation and chemotherapy. Although there are several reports on genetic mutations epigenetic changes define the pathogenesis of MPNST, insufficient information regarding microenvironment contributes malignancy MPNST. In present study, we demonstrate adrenaline increases cancer stem cell population This effect mediated by stimulation beta-2 adrenergic receptor (ADRB2), which activates Hippo transducer, YAP/TAZ. Inhibition RNAi experiments revealed inhibition ADRB2 attenuated adrenaline-triggered activity YAP/TAZ subsequently MPNST cells stemness. Furthermore, ADRB2-YAP/TAZ axis was confirmed patients’ specimens. The prognosis patients with high levels found significantly worse.
